RESUMO: 

Hepatotoxicidade de plantas medicinais. XXXIII A&#xE7;&#xE3;o de Tribulus terrestris L. em ratos.Tribulus
terrestris L. (fam&#xED;lia Zygophyllaceae), comumente conhecida como estrepe, trepadeira-perfurante e cabe&#xE7;ade-gato, 
&#xE9; uma erva anual procumbente, largamente distribu&#xED;da no Sul da Europa, Austr&#xE1;lia, Am&#xE9;rica,
&#xC1;frica e &#xC1;sia, h&#xE1; muito utilizada na medicina chinesa e indiana no tratamento de v&#xE1;rias doen&#xE7;as e &#xE9;
popularmente clamada por melhorar a fun&#xE7;&#xE3;o sexual no homem; &#xE9; utilizada pelos atletas para promover
a hipertrofia muscular. A planta foi utilizada com o objetivo de estudar seus efeitos nas estruturas do f&#xED;gado
de rato. T. terrestris (sementes e frutos) foi administrada oralmente via sonda g&#xE1;strica na dosagem de 5
mg, uma vez ao dia, durante 60 dias. Todos os ratos foram sacrificados e o f&#xED;gado fixado em formol a 10%.
Os cortes de 6 pm de espessura foram corados com hematoxilina e eosina. Foram utilizadas t&#xE9;cnicas
cariom&#xE9;tricas e estereol&#xF3;gicas. A an&#xE1;lise histopatol&#xF3;gica revelou &#xE1;reas periportais, intermedi&#xE1;rias e
pericentrolobulares com hepat&#xF3;citos apresentando v&#xE1;rios graus de vacuoliza&#xE7;&#xE3;o. Muitos hepat&#xF3;citos est&#xE3;o
balonizados. A veia centrolobular est&#xE1; dilatada e congesta. Foram observados sinusoides dilatados
contendo eritr&#xF3;citos. Les&#xF5;es inflamat&#xF3;rias focais ocorreram no par&#xEA;nquima e &#xE1;reas portais dos animais
tratados. O espa&#xE7;o-porta mostrou fibrose, prolifera&#xE7;&#xE3;o do dueto biliar e colangi&#xF3;citos pequenos. O estudo
histom&#xE9;trico mostrou volumes citoplasm&#xE1;tico e celular menores, e valores aumentados para o n&#xFA;mero
de hepat&#xF3;citos por mm.3. Os n&#xFA;cleos dos hepat&#xF3;citos e colangi&#xF3;citos eram menos volumosos. Estes
resultados indicam que a planta cont&#xE9;m um ou mais constituintes t&#xF3;xicos, provavelmente saponinas
esteroidais, que provocam dano hep&#xE1;tico que poderia explicar a perda de peso corporal do rato.

ABSTRACT: 

Tribulus terrestris L. (family Zygophyllaceae), commonly known as caltrop, puncture vine and
cat-head, is a prostate annual herb, widely distributed in South Europe, Australia, America, Africa and Asia,
has long been used in traditional Chinese and Indian medicine for the treatment of various illnesses and
is popularly claimed to improve the sexual functions in man; it is used by athletes to promote muscular
hypertrophy. The herb was used to study its effects on rat liver structures. T. terrestris (seeds and fruits) was
orally administered via gavage in the dose of 5 mg, once daily for 60 days. All rats were sacrificed and liver
was fixed in 10% formaline. The 6 pro-sections were stained with H &amp; E. Karyometric and stereologic
techniques were employed. Histopathological analysis revealed periportal, midzonal and pericentrilobular
regions with parenchymal cells with varying degrees of vacuolation. Many hepatocytes are ballooned. The
centrilobular vein was dilated and congested. Dilated sinusoids containing erythrocytes were observed.
Focal inflammatory lesions occurred in treated animals parenchyma and portal area. The portal area
showed fibrosis, biliar duct proliferation with small cells. Histometric study showed smaller cytoplasm and
cell volumes, and higher values for number of hepatocytes per mm3. The hepatocytes and cholangiocytes
nuclei were less voluminous. These results indicate that the herb contains one or more toxic constituents,
probably steroidal saponins, and damage of liver could explain the loss of body weight.
