RESUMO: 

Popularmente conhecida como cabeludinha, a Plinia glomerata (O. Berg) Amshoff
&#xE9; uma esp&#xE9;cie nativa e t&#xED;pica da regi&#xE3;o da Mata Atl&#xE2;ntica. Como a planta ainda encontra-se
em fase de domestica&#xE7;&#xE3;o, estudos relacionados &#xE0; propaga&#xE7;&#xE3;o e &#xE0; sele&#xE7;&#xE3;o de gen&#xF3;tipos
promissores para a propaga&#xE7;&#xE3;o s&#xE3;o necess&#xE1;rios. O cultivo in vitro &#xE9; uma importante
ferramenta em programas de conserva&#xE7;&#xE3;o de recursos gen&#xE9;ticos e melhoramento gen&#xE9;tico.
Considerando esses aspectos, no presente estudo objetivou-se avaliar a germina&#xE7;&#xE3;o in vitro de
sementes de P. glomerata utilizando-se diferentes concentra&#xE7;&#xF5;es de &#xE1;cido giber&#xE9;lico (GA3) e a
organog&#xEA;nese in vitro a partir de explantes foliares submetidos a diferentes concentra&#xE7;&#xF5;es de
6-benzilaminopurina (BAP) combinado com &#xE1;cido naftalenoac&#xE9;tico (ANA). Para a germina&#xE7;&#xE3;o
in vitro, foram realizados dois experimentos: 1) as sementes foram primeiramente embebidas
por 24 h em &#xE1;gua est&#xE9;ril antes da assepsia com hipoclorito de s&#xF3;dio e 2) as sementes foram
primeiramente desinfestadas e depois embebidas por 24 h em &#xE1;gua est&#xE9;ril. Para a germina&#xE7;&#xE3;o
in vitro, utilizou-se o meio de cultura MS suplementado com diferentes concentra&#xE7;&#xF5;es de GA3:
0,0; 0,5; 1,0; 2,0; 5,0 e 10,0 mg L-1 de GA3. Para a organog&#xEA;nese, utilizou-se explantes foliares
oriundos de pl&#xE2;ntulas germinadas in vitro, sendo inoculados em meio MS suplementado com
diferentes concentra&#xE7;&#xF5;es de BAP (0; 0,1; 0,25; 0,5; 1,0 e 2,0 mg L-1) combinadas com 0,1
mg L-1 de ANA. O procedimento de assepsia foi realizado somente ap&#xF3;s as sementes serem
embebidas em &#xE1;gua destilada est&#xE9;ril. Para porcentagem de germina&#xE7;&#xE3;o in vitro, observou-se
um incremento na germina&#xE7;&#xE3;o proporcional ao aumento da concentra&#xE7;&#xE3;o, variando entre 2,5%
no tratamento controle e 85% para a concentra&#xE7;&#xE3;o de 10 mg L-1 de GA3, demostrando que as
sementes de P. glomerata necessitam de aplica&#xE7;&#xE3;o ex&#xF3;gena desse regulador vegetal para
otimizar a germina&#xE7;&#xE3;o in vitro. Na organog&#xEA;nese, quando utilizou-se 1,0 mg L-1 de BAP e 0,1
mg L-1 de ANA, 92% de explantes apresentaram calos.

ABSTRACT: 

Germination and organogenesis in vitro of Plinia glomerata (O. Berg) Amshoff
. Popularly known as cabeludinha, P. glomerata (O. Berg) Amshoff is a native and typical species
of the Atlantic Forest region. Still in the domestication phase, some aspects of propagation and
selection of promising genotypes for an in vitro propagation are needed, and the development
of in vitro culture protocols is very important for genetic resources maintenance programs and
for genetical enhancement. This study aimed to perform the in vitro germination of seeds P.
glomerata using different concentrations of gibberellic acid (GA3), and the in vitro organogenesis
of leaf explants with different BAP (6-benzilaminopurine) concentrations combined with ANA
(naphtaloacetic acid). For in vitro germination, two experiments were conducted: 1) seeds
were first embedded for 24 h in sterile water prior to sodium hypochlorite asepsis and 2) seeds
were first sterilized and then embedded for 24 h in sterile water. It was used MS medium for
germination in vitro with different concentrations of GA3: control treatment without addition
of plant growth regulator; 0.5, 1.0, 2.0, 5.0 and 10.0 mg L-1 of GA3. During organogenesis,
seedling leaves were germinated in vitro. The explants were inoculated in MS six treatments:
0, 0.1, 0.25, 0.5, 1.0 and 2.0 mg L-1 of BAP, all combined with 0.1 mg L-1 of NAA. After 30 days
of in vitro culture, the seeds of the first experiment presented tissue of primary root and shoot.
However, there was no germination in treatments with GA3 at the second experiment. For the 
variable in vitro germination percentage was observed an increase in germination proportional
to the increase in concentration ranging from 2.5% in the control treatment and 85% in the
concentration 10 mg L-1 of GA3, showing that P. glomerata seeds require exogenous application
of growth regulator to the higher percentage germination occurs. During organogenesis, 92%
of explants presented callus.
