RESUMO:    Marcadores moleculares permitem acompanhar o progresso de um programa de  melhoramento  gen&#xE9;tico.  Marcadores  SSR  (simple  sequence  repeats),  por  serem  codominantes, multial&#xE9;licos e apresentarem alto polimorfismo, s&#xE3;o ideais para estudos de cria&#xE7;&#xE3;o de mapas g&#xEA;nicos, variabilidade e confirma&#xE7;&#xE3;o de retrocruzamentos. Por&#xE9;m, o uso deste tipo de marcador molecular &#xE9; limitado, visto que s&#xE3;o apenas dispon&#xED;veis primers para esp&#xE9;cies com express&#xE3;o comercial. Neste contexto, o objetivo deste trabalho foi avaliar a transferibilidade de 21 loci microssat&#xE9;lites desenvolvidos e desenhados de P.edulis e P. alatapara quatro esp&#xE9;cies silvestres de Passiflora. As esp&#xE9;cies utilizadas foram: P. cincinnata, P.   quadrangularis, P. mucronata, e P. nitida. A extra&#xE7;&#xE3;o de DNA foi realizada atrav&#xE9;s do Kit Promega Wizard&#xAE;. O protocolo de amplifica&#xE7;&#xE3;o utilizado foi adaptado do proposto para o maracujazeiro comercial. Foram utilizados dezenove marcadores microssat&#xE9;lites desenhados para  a  esp&#xE9;cie  P.  edulis  e  dois  desenhados  para  P.alata. Os amplicons foram levados &#xE0; eletroforese horizontal em gel de agarose a 3% e corados com o buffer Nancy 520. Os g&#xE9;is foram revelados em fotodocumentador digital e analisados. A transferibilidade foi verificada atrav&#xE9;s da avalia&#xE7;&#xE3;o de presen&#xE7;a ou aus&#xEA;ncia de banda. Os loci microssat&#xE9;lites projetados para P. edulis e P. alata mostraram-se transfer&#xED;veis para ao menos uma das esp&#xE9;cies com uma taxa altamente vari&#xE1;vel sendo, P. cincinnata (33,33%), P. quadrangularis (23,81%), P.   mucronata (19,05%) e P. nitida (14,28%). Os marcadores PE13 e PE54 podem ser indicados para estudos de caracteriza&#xE7;&#xE3;o molecular por serem os &#xFA;nicos capazes de amplificar todas as esp&#xE9;cies de maracuj&#xE1; silvestre estudadas.

ABSTRACT:    Molecular markers are tools that allow the monitoring of a breeding program progress. SSR (simple sequence repeats) markers are applied for genetic mapping studies, variability,  assisted  selection,  crossing  and  backcrossing,  because  they  are  codominant,  multiallelic  and  high  polymorphic.  However,  the  use  of  this  type  of  molecular  marker  is  limited,  since  primers  are  only  available  for  commercially  expressed  species.  Therefore,  this  work  aimed  to  evaluate  the  transferability  designed  primers  of  Passiflora  alata  e  Passiflora edulis to four wild species of Passiflora. The species used were: P. cincinnata, P. quadrangularis, P. mucronata and P. nitida. DNA extraction was done through the Promega Wizard&#xAE; Kit. The amplification protocol used was the one proposed for P. alata and P. edulis with adaptations. Nineteen microsatellites designed for P. edulis and two designed for a genus P. alata were used. The amplicons were taken to horizontal electrophoresis in a 3% agarose gel, followed by staining with Nancy 520 buffer and the revelation was developed in a digital photodocumentator with UV light. The transferability was verified through the evaluation of band presence or absence. The microsatellites loci designed for P. edulis and P. alata  were  transferable  for  the  examination  of  at  least  one  species  with  high  variable  rate: P. quadrangularis (23.81%), P. cincinnata (33.33%), P. quadrangularis (23.81%), P. mucronata (19.05%) and P. nitida (14.28%). Markers PE13 and PE54 can be selected for molecular characterization studies because they are the only ones able to amplify all the wild passion fruit species tested.
